摘　　要：目的研究orexinA对麻醉大鼠前额叶皮层神经元自发放电的影响,并初步探讨其作用机制。方法用玻璃微电极记录大鼠前额叶皮层神经元的单位放电,分别观察侧脑室给予orexinA、组胺H1受体拮抗剂pyrilamine、以及pyrilamine+orexinA后放电活动的变化。结果侧脑室给orexinA溶液可明显增加前额叶皮层神经元的自发放电频率;而给pyrilamine对前额叶皮层神经元自发放电频率无显著影响;给予pyrilamine预处理后,orexinA未能引起前额叶皮层神经元放电频率发生改变。结论OrexinA对麻醉大鼠前额叶皮层神经元放电活动有明显的兴奋性,这一作用可能与活跃的组胺能系统有关。Objective To study the effect of orexin A on the spontaneous unit discharges of the prefrontal cortical neurons in anesthetized rats. Methods The unit discharges of neurons in the prefrontal cortex were recorded by glass microelectrode technique before and after intracerebroventricular injection of orexin A, pyrilamine （a histamine H1 receptor antagonist）, or pyrilamine ＋ orexin A. Results Extracellular single-unit recordings were obtained from a total of 55 prefrontal cortical neurons. Orexin A significantly induced the increase of the discharge rate. But no changes of discharge rate were observed after administration of pyrilamine. In contrast, orexin A failed to exert an excitatory action on unit discharges of cortical neurons following pretreatment with pyrilamine in the lateral ventricle. Conclusion Orexin A exerts an excitatory action on prefrontal cortical neurons, which probably results from activation of histamine H1 receptor pathways.
