摘 要:目的探讨Aβ亚单位疫苗诱发体液免疫反应的量效关系和时程。方法采用C57BL小鼠,依据滴入Aβ15的量将其分为50、25、12.5μg3个组,每周1次经鼻粘膜滴入Aβ15亚单位疫苗,观察1年之中鼠血清中抗Aβ抗体水平的变化;取脾细胞培养,加入不同浓度的Aβ15,用MTT法观察脾细胞增殖情况。结果3组小鼠的外周血内均有抗Aβ抗体产生。25μg组和12.5μg组产生抗体较50μg组早,3组峰浓度抗体水平分别为1∶6720、1∶6222和1∶3565。脾细胞增殖实验表明0.5μg和5μg的Aβ15具有较强的刺激脾细胞增殖的作用,而50μg的Aβ15对脾细胞增殖的刺激作用反而下降(P=0.042;P=0.021)。结论Aβ15亚单位疫苗诱导体液免疫反应的能力因接种的剂量不同而异。高浓度的Aβ15可以引起免疫耐受,使抗Aβ抗体产生减少。Aim To investigate the duration of humoral immune reaction which may conduce to understand the immune mechanism of Aβ subunit vaccine and determine the optimal dose and time course. Methods C57BL mice were divided into three groups, 50, 25 and 12.5 μg, according to the immune doses. Immune via nasal mucous membrane weekly and employ indirect ELISA assay to detect the anti-Aβ antibodies level in the serum. Splenocytes were cultured with different concentration Aβ15 ,0.5 mg·L^-1, 5 mg· L^-1 and 50 mg · L^-1. Cells proliferation rate was measured using MTT assay. Results The anti-Aβ antibodies were detected in 3 groups, they were emerged earlier in 25 μg and 12.5 μg groups than the 50 μg group. Moreover, the peak concentrations in three groups were 1 : 6 720,1 : 6 222 and 1 : 3565. The splenocyte proliferation assay showed that the higher A value were found in the group of 0.5 mg· L^-1 and 5 mg · L^-1 Aβ15 than the 50 mg · L^-1 group(P = 0. 042 ;P = 0. 021, respectively). Conclusion Aβ15 subunit vaccine induces the humoral immune reaction dependent of the dose inoculated. High level Aβ15 also induces the immunologic tolerance and leads to the decrease of anti-Aβ antibodies.
