家蝇抗菌物质超声波诱导效果的动态观察

摘　　要：目的观察超声波诱导家蝇抗菌物质的动态表达效果。方法家蝇3龄幼虫经100 W超声波处理5 min后,分别于第0、24、487、2 h观察、记录幼虫存活率;提取其血淋巴,以溶壁微球菌作指示菌,采用平板法作抑菌试验,37℃恒温24 h,观察抑菌圈,以抑菌圈直径表示抗菌活力的大小,借以指示家蝇抗菌物质诱导表达的动态规律。结果经超声波诱导后,第24、487、2 h幼虫存活率依次为(13.41±9.19)%(、10.90±8.06)%和(6.82±4.93)%,第0、24、48、72 h收集的血淋巴对溶壁微球菌的抑菌环直径依次为(13.00±1.00)、(21.83±0.58)(、22.33±0.76)和(22.75±0.35)mm。结论超声波处理可诱导家蝇产生抗菌物质,在诱导后第48 h收集血淋巴最为合适。Objective To investigate the dynamics of the antibacterial activity against Micrococcus lysodeikticus in the housefly Musca domestica after being induced by ultrasonic. Methods The third instar larvae were induced by ultrasonic waves at 100 watts for 5 min, survival rates of the treated larvae were recorded at different hours after treatment, and hemolymph collected at hours 0, 24, 48, 72 was tested against M. lysodeikticus respectively, the bacteriostatic effects were observed and diameters of clear inhibitory ring were recorded to express antibacterial activity. Results After being treated for 24, 48, 72 h, the larval survival rates were orderly recorded as follows： （13.41±9.19）%, （10.90±8. 06）%, （6.82±4.93） %. After treatment for 0, 24, 48, 72 h, the diameters（mm） of antibacterial rings of the hemolymph were 13.00±1.00, 21.83±0.58, 22.33±0.76, 22.75±0.35 respectlvely. Conclusion The ultrasonic treatment can induce antibacterial substance in the housefly, the hemolymph should be collected after being treated for 48 h.
