摘  要:目的运用血清药理学方法,体外研究温下方对A549/DDP细胞多药耐药的逆转作用及机制。方法正常血清及不同浓度的温下方含药血清作用于A549/DDP细胞,四甲基偶氮唑蓝(MTT)法检测温下方含药血清对A549/DDP细胞的逆转作用;运用免疫荧光技术,激光共聚焦显微镜及流式细胞仪检测肺耐药相关蛋白(LRP)、多药耐药相关蛋白(MRP)和P-糖蛋白(P-gp)的表达。结果温下方含药血清能明显逆转A549/DDP细胞的多药耐药,增敏倍数为2~2.5倍。并可明显降低P-gp、LRP、MRP蛋白表达。结论温下方通过降低P-gp、LRP、MRP的表达以增强A549/DDP对化疗药的敏感性,逆转肺腺癌细胞的多药耐药。Objective To identify the effect and underlying mechanism of Wenxiafang,an empirical herbal formula of traditional Chinese,in regression of multiple chemotherapeutic resistance in A549/DDP ceils,using serum pharmacological methods in vivo. Methods A549/DDP cells were incubated with normal mouse serum or serum containing varied dose levels of Wenxifang,then assayed by MTY for regression of multiple chemotherapeutic resistance. The expressions of lung resistance-related protein (LRP), multidrug resistance associated protein (MRP) and P-glycoprotein (P-gp) in cells were evaluated by immunofluorescence technique,laser scanning confocal microscopy and flow cytometry. Results Serum containing varied dose levels of Wenxifang significantly regressed multiple chemotherapeutic resistance in A549/DDP cells by a 2-to-2.5-fold increase in sensitivity,and markedly suppressed the expressions of P-gp,LRP and MRP in the cells. Conclusion Wenxiafang appeared to increase the chemotherapeutic sensitivity of A549/DDP cell by down-regulating expressions of P-gp,LRP and MRP,thereby showing regression of multiple chemotherapeutic resistance in pulmonary adenocarcinoma.
