摘 要:目的研究甲状腺乳头状癌组织中人血小板第IV因子(hPF4)表达与微血管密度(MVD)、淋巴结转移之间的关系。方法对40例甲状腺乳头状癌手术标本和10例癌旁正常组织标本采用反转录聚合酶链反应(RT-PCR)检测hPF4mRNA表达,并采用CD105单克隆抗体标记免疫组化法检测MVD。结果正常组织hPF4mRNA阳性率为80%;癌组织hPF4阳性率为47.5%,表达组MVD(25.4±11.7)低于不表达组(45.1±11.9),MVD差异有统计学意义(t值为2.64,均P<0.01),hPF4mRNA阳性表达率在发生淋巴结转移的病例显著低于没发生转移的病例(χ2=9.42,P<0.01)。结论甲状腺乳头状癌组织hPF4mRNA表达可降低MVD和抑制淋巴结转移。Objective To study the expression of human platelet factor 4 (hPF4)mRNA and its relationship with angiogenesis and lymph node metastasis in papillary throid carcinoma (PTC). Methods RTPCR was used to detect the expression of hPF4 mRNA in 40 cases of PTC and 10 cases of normal tissue surrounding carcinoma. Immunohistochemical stain was performed to detect microvessel density(MVD) whose endothelia were marked by antibody CD105. Results The positive-expression rate of hPF4 mRNA was 80 % respectively in normal tissue surrounding carcinoma. The positive-expression rate of hPF4 was 47.5 % in PTC cases. The MVD value of PTC cases with hPF4 mRNA positive expression was significantly lower than that of cases without hPF4 mRNA expression (P 〈 0.01). The expression rates of hPF4 mRNA of PTC cases with lymph node metastasis were significantly lower than those without metastasis (X^2 = 9.42, P 〈 0.01). Conclusion The expression of hPF4 mRNA may decrease MVD and prohibit lymph node metastasis.
