溃疡性结肠炎与结肠癌肠黏膜中p27蛋白和PCNA的表达及凋亡的分析

摘　　要：目的探讨p27蛋白和增殖细胞核抗原(PCNA)在溃疡性结肠炎、结肠癌肠黏膜中的表达及其与细胞凋亡的关系。方法应用免疫组化SABC方法检测结肠癌、溃疡性结肠炎患者及正常人结肠肠黏膜中p27蛋白和PCNA的表达,用DNA原位末端标记TUNEL法检测标本中的细胞凋亡。结果结肠癌及溃疡性结肠炎组织中的PCNA表达水平明显高于正常结肠(P<0.05);p27蛋白阳性表达的结肠癌细胞平均凋亡指数(0.93±0.32)%高于p27蛋白阴性表达的细胞凋亡指数(0.39±0.18)%,差异有统计学意义(P<0.05),p27与结肠癌PCNA的表达无关。结论溃疡性结肠炎结肠黏膜标本PCNA表达呈强阳性且组织分布弥散提示结肠黏膜癌或有癌变倾向;p27蛋白表达可能不抑制结肠癌细胞的增殖,但有促使结肠癌细胞凋亡的趋势。Objective To analyze the expressions of p27 and PCNA and cell apoptosis in ulcerative colitis and colon cancer. Methods The expressions of p27 and proliferation cell nuclear antigen（PCNA） proteins in colorectal cancer, ulcerative colitis and normal colon were analyzed by StrptAvidin-Biotin Enzyme Complex immunohistochemical methods. The apoptosis was detected using terminal uridine deoxynucleiotide nick end labeling（TUNEL） histochemistry. Results The level of PCNA expression in colorectal cancer and ulcerative colitis was markedly higher than that in normal colon（ P 〈 0. 05 ）. The MAI in p27 positive patients （0. 93 ± 0. 32） was higher than that of p27 negative ones（0. 39 ±0. 18） in colorectal cancers（ P 〈0. 05）. The expression of PCNA in colon cancer has no relationship to p27. Conclusions The high expression of PCNA and the diffusive pattern of tissue distribution in ulcerative colitis may indicate cancer or a tendency to become cancer. The expression of p27 may not inhibit the cellular proliferation, but may accelerate apoptosis of colorectal cancer cells.
