重组真菌细胞色素P450nor2在酵母细胞中的表达

摘　　要：酿酒酵母(Saccharomyces cerevisiae)是一种理想的真核蛋白表达系统.将真菌细胞色素P450nor2基因亚克隆到酵母表达载体pAUR123中,构建重组表达质粒pAUR-P450nor2并转化酿酒酵母AH22,经Aureobasidin A筛选和菌落PCR鉴定得到阳性克隆.SDS-PAGE分析证实:重组的真菌细胞色素P450nor2在酵母细胞中实现了高表达.Saccharomyces cerevisiae was a desired expression system for eukaryotic protein. Cytochrome P450nor gene was subcloned into the shuttle vector pAUR123 to yield the recombinant plasmid pAUR-P450nor2. The recombinant plasmid pAUR-P450nor2 was transformed into S. cerevisiae AH22 and got positive clone after the screening with Aureobasidin A and colony PCR. The SDS-PAGE showed that the expression product was recombinant cytochrome P450nor2 and got a high level.
