摘　　要：目的:观察胃泌素对人低分化胃癌细胞株BGC-823的生长调控作用,测定细胞内环磷酸腺苷(cAMP)含量、蛋白激酶C(PKC)活性及PKC亚型的表达,探讨受体后信息传导途径。方法:应用MTT法观察细胞的增殖程度,放射免疫分析方法测定细胞内cAMP含量[,-γ32P]ATP掺入外源性底物的方法测定PKC活性,Western blot方法分析PKC亚型α、1β、2β及ε的表达。结果:胃泌素能促进BGC-823细胞的生长,且与剂量呈正相关(r=0.74,P<0.05);这种促生长作用被胃泌素受体拮抗剂丙谷胺拮抗。胃泌素能促进细胞PKC活性的增加c,AMP含量无明显变化;胃泌素使PKCα表达明显增加,PKC1β、PKC2β及PKCε无明显表达。结论:胃泌素促进胃癌细胞BGC-823生长的作用由相应的受体介导,其受体后信息传递途径可能涉及二酯酰甘油蛋白激酶C系统,PKCα在胃泌素的受体后信息传递中起重要作用。Objective： To observe the effect of gash-in on the growth of poorly differentiated human gastric cancer cell line BGC-823 and detenTtine the level of intracellular cyclic adenosine monophosphate （cAMP） ,the activity of protein kirnase C （PKC） ,and the expression of PKC isoforms, PKC-α, PKC-β1, PKC-β2, and PKC-ε, and to explore the post-receptor signal transduction mediated by gastrin. Methods： The proliferation of BGC-823 cells, the level of intracellular cAMP, the activity of PKC, and the expression of PKC isoforms were detemained by MTF assay,radioimmunoassay,incorporation of [γ-^32P ]-ATP into exogenous substrate, and Western blot,respectively. Results： Gasults：Gastrin promoted the growth of BGC-823 cells in a dose-dependent manner（r =0.74, P 〈 0.05）,and this effect of gastrin was significantly inhibited by proglumide, the antagonist of gastrin. Gasu＇in increased the activity of PKC and the expression of PKC-of.PKC-α Conclusion： Gasu＇in could promote the growth of human gastric cancer cell line BGC-823 via related receptor,in which diacylglycerol-PKC may be involved. PKC-α may play an important role in the pest-receptor signal transduction mediated by gastrin.
