摘  要:目的:探讨低硒饮食对发育期大鼠大脑皮层和海马的脂质过氧化物和转录因子cAMP反应元件结合蛋白(CREB)的影响。方法:生化分析方法测定低硒喂养的第三代21日龄大鼠不同脑区丙二醛(MalondialdehydeMDA)含量的变化,用免疫组织化学和Westernblot免疫印迹的方法测定CREB及磷酸化CREB(p-CREB)的含量。结果:低硒饮食组较对照组大鼠大脑皮层和海马MDA含量增加,CREB和p-CREB含量下降。结论:低硒膳食能够引起发育期大鼠大脑皮层和海马的氧化代谢产物堆积。CREB表达减少,提示低硒能够引起大脑皮层和海马神经元功能下降,低硒可能是导致学习记忆能力降低的潜在因素之一。Object: To observe the changes of lipid peroxide and cAMP Response Element-Binding Protein (CREB) in the cortex and hippocampus of developing rat induced by selenium deficiency diet. Material and method: The dissected cortex and hippocampus of the third generation 21-day rats were assayed by 2-thiobarbituric acid (TBA) test to determine the contents of MDA. The expression of CREB and p-CREB were examined by Western blot and immunohistochemistry. Result: The content of MDA in the cortex and hippocampus of selenium-deficient group was much higher than that of the control group. The detection of CREB and p-CREB showed that the expressions of selenium-deficient group were lower than that of control group. Conclusion: (1) The decline of oxidant metabolism in the developing cortex and hippocampus can be induced by selenium deficiency diet. (2) The reduction of the expression of CREB and p-CREB reflected the decreased cortex and hippocampus neuron functions. It is indicated that selenium deficiency is one of the possible reasons to decline the learning and memory ability.
