摘 要:目的探讨不同条件低强度氦氖激光照射对体外培养的兔角膜内皮细胞增生的影响。方法体外培养的兔角膜内皮细胞经低强度激光照射后,MTT比色法检测其增生活性;通过生物显微镜、组织化学染色观察其照射前后形态学变化;通过流式细胞仪检测其照射前后细胞周期的变化等。结果MTT检测结果显示,体外培养的兔角膜内皮细胞经低强度激光照射后,其增生活性提高,本实验中最适照射强度和时间分别是5mW·cm-2、600s;流式细胞仪细胞周期检测结果显示:激光照射组的细胞增生指数明显高于对照组,而与碱性成纤维细胞生长因子组比较差别无显著性意义。结论低强度激光在一定程度上可促进角膜内皮细胞的增生。Objective To investigate the effects of different low-intensity Hc-Ne laser irradiation on proliferation of cultured rabbit corneal endothelial ceils(CECs) in vitro. Methods After irradiated by low-intensity laser,the prolfferative activity of CECs were assessed by MTT assay. The changes of ceil cycle before and after laser irradiated were examined by flow cytometry. The morphological changes of CECs before and after laser irradiated were observed by biological microscope and histochemical stain. Results The restdts of MTT assay indicated that the proliferative activity of cultured rabbit CECs in vitro was increased after laser irradiation and culturing, the CECs grew rapidly and increased quantitatively. The optimal laser irradiation energy intensity and time in this experiment was 5 mW·cm^-2 ,600 s. The results of flow cytometry showed that the proliferative index in laser irradiated group was much higher than that in the control group,but there was no difference compared with the buck flbroblast growth factor group. Conclusion Low-intensity laser can promote the proliferation of cultured rabbit in vitro in certain extent.
