摘  要:目的研究雷公藤内酯醇(triptolid,T10)对血管内皮细胞移行活性及组织型纤溶酶原激活物(tissue-typeplasminogenactivator,t-PA)和纤溶酶原激活物抑制物-1(plas-minogenactivatorinhibitor-1,PAI-1)蛋白表达的影响,探讨T10对新生血管生成的抑制作用。方法体外培养传3代人脐静脉内皮细胞(humanumbilicalveinendothelialcells,HUVECs),加入不同浓度的T10(5μg·L-1、10μg·L-1、20μg·L-1、30μg·L-1)、地塞米松(300mg·L-1),观察HUVECs移行活性;免疫组织化学染色检测HUVECs中t-PA和PAI-1蛋白表达量。结果T10能明显抑制HUVECs移行活性,使HUVECs移行数量、移行距离明显减少和缩短;T10能明显抑制HUVECs中t-PA和PAI-1蛋白表达,使t-PA和PAI-1蛋白表达棕黄色阳性染色减少,与对照组相比差异具有显著性(P<0.01)。结论T10能明显抑制血管内皮细胞移行,抑制内皮细胞t-PA和PAI-1蛋白表达,从而有效抑制新生血管生成与生长。Objective To observe the effects of trtptolide on the migration activity,protein expression of tissue-type plasminogen activator(t-PA) and. plasminogen activator inhibitor-1 ( PAI-1 ) of human umbilical vein endothelial cells (HUVECs) cultured in vitro, and elucidate the anti-angiogenic mechanism of triptolide. Methods HUVECs were cultured in vitro and the third passage cells were treated with triptolide (5μg · L^-1,10μg · L^-1,20μg · L^-1,30μg · L^-1) ,or dexamethasone ( 300 mg·L^-1 ) as control. The effects of triptolide on the migration of HUVECs were observed,and immunohistochemical assay was applied to detect protein expression of t-PA and PAI-1. Results Triptolide inhibited the migration activity obviously, shortened the migrated distance and reduced the number of HUVECs. Triptolide suppresed the protein expression of t-PA and PAI-1 ,and reduced the brown positive staining. There were obvious differences compared with the control group(P 〈0. 01 ). Conclusion Triptolide can inhibit the migration activity of HUVECs,and down-regulate protein expression of t-PA and PAI-1 ,so it can inhibit the angiogenesis effectively.
