摘 要:目的探讨2种糖尿病大鼠模型血-视网膜屏障对伊凡思蓝(Evans-Blue,EB)通透性的改变。方法对照组及1周、4周和16周时Sprague-Dawley(SD)和Brown-Norway(BN)糖尿病大鼠,EB45mg·kg-1颈静脉注入,分别检测视网膜中EB含量。结果对照组及1周、4周和16周时SD和BN糖尿病大鼠视网膜标准化EB含量分别为(12.24±3·11)ng·mg-1、(21.68±4.41)ng·mg-1、(21.78±3.17)ng·mg-1、(23.56±4.05)ng·mg-1和(13.22±3.59)ng·mg-1、(23.41±4.47)ng·mg-1、(26.40±5.00)ng·mg-1、(31.01±4.46)ng·mg-1,糖尿病大鼠视网膜标准化EB含量明显高于对照组,其差异均有显著性(P<0·05)。2种不同种系大鼠相比,BN糖尿病大鼠1周、4周和16周时视网膜标准化EB含量均显著高于SD大鼠(P<0.05),而对照组差异无显著性(P>0.05)。结论通过检测视网膜内EB含量,可以比较精确的发现早期糖尿病大鼠视网膜病变中血-视网膜通透性的改变。不同大鼠种系的差异性决定了其对STZ诱导的糖尿病大鼠视网膜通透性改变的不同。Objective To investigate the changes of two species diabetic rats animal model for Evans-Blue (EB) permeability to the blood-retina barrier. Methods After EB(45 mg · kg^-1 )was injected throttgh jugular vein,control group and 1 week, 4 weeks and 16 weeks diabetic Sprague-Dawley (SD)and Brown-Norway(BN) rats were respectively detected for the content of EB in the retina. Results Retinal EB leakages of retina from control, group 1 week, 4 weeks, and 16 weeks for two species diabetic rats were ( 12.24±3. 11 )ng ·mg^-1, (21.68±4.41)ng ·mg^-1 ,(21.78±3.17 )ng ·mg^-1,(23.56 ±4.05 )ng ·mg^-1 and (13.22±3.59)ng ·mg^-1,(23.41±4.47 )ng ·mg^-1,(26.40±5.00)ng ·mg^-1, ( 31.01±4.46)ng ·mg^-1,respectively. EB in diabetic rats was much higher than that in control group( P 〈 0.05 ). Compared the rats of two different breeds, EB of BN diabetic rats was much higher than that of SD rats( P 〈0.05 ), which was detected in 1 week,4 weeks and 16 weeks,and there was no significant difference compared with the control group ( P 〉 0. 05 ). Conclusion By detecting EB, the change of blood-retinal barrier permeability of diabetic rats with retinal lesion in early stage can be exactly observed. Genetic background determines the changes of retinal permeability in diabetic rats induced by SIZ.
