Laminin五肽对胃癌细胞系MGc－803的体外抑制作用

摘　　要：描述了Ｌａｍｉｎｉｎ五肽（ＹＩＧＳＲ）对胃癌细胞系ＭＧｃ－８０３的体外抑制作用，在多肽浓度为１ｍｇ／ｍｌ时抑制率高达７３％．抑制作用的机理可能是Ｌａｍｉｎｉｎ五肽竞争了肿瘤细胞上Ｌａｍｉｎｉｎ受体，从而封闭了肿瘤细胞对基底膜的结合及增殖．因此，利用Ｌａｍｉｎｉｎ五肽一类的药物有可能为肿瘤术后防止扩散提供一个新方法．The inhibition of laminin pentapeptide (YIGSR) on gastric cancer cell line MGc-803 in vitro was described. Cell cultures were set up in flat-bottomed 96-well culture trays,and each well was of a volume of 100 al containing 5×105 cells/ml. Then different concentrations of peptide and 3H-thymidine were added to the cultures and incubated at 37 ℃ for 3 days in a humidified incubator with 5% CO2. The inhibition rates of l mg/ml,0. 1 mg/ml and 0. 01 mg/ml of peptide were 73%,52% and 35%,respectively.The results indicated that laminin pentapeptide is a strong inhibitor of tumor cells. A possible mechanism of its inhibition has been suggested.
