摘　　要：①目的探讨bcl-2基因转染对人胃上皮细胞GES-1细胞周期的影响。②方法采用携带新霉素抗性(NEOr)基因的pcD-NA3/bcl-2和脂质体(lipofectamine)法转染人胃上皮细胞系GES-1,同时以pcDNA3(NEOr)空载体质粒转染同一细胞作为对照,用新霉素418(G418)筛选出bcl-2转染的阳性克隆。流式细胞术检测各组细胞细胞周期。③结果bcl-2基因转染组S期细胞数明显较对照组增多。④结论bcl-2基因转染可使S期细胞增多,提高转染细胞的增殖能力。Objective To test the effect of bcl- 2 gene transfection on the cell cycle of human gastric epithehal cen line GES - 1. Methods Lipofectamine method was adopted to transfect PcDNA3/bcl -2 into human gastric epithehal cell line GES - 1. The GES - 1 transfected by empty vector peDNA3 carrying NEOr were used as control. The geneticin 418 （G418） was adopted to select positive clones of bcl - 2 gene transfected cens. The cen cycle of each group cens was tested by flow eytometry （FCM）. Results The results of FCM analysis showed that the proportion of S - phase cells in bcl - 2 gene transfected group increased obvi- ously. Conclusion bcl - 2 gene transfection can increase the proportion of S - phase cells and enhance the proliferation ability of bcl- 2 gene transfected ceils.
