摘 要:目的观察神经再生素(NRF)影响海马神经元生长的机制。方法用胚胎17d的大鼠海马离体培养神经原,分用药组和对照组,培养6h加NRF(1μg/ml),检测神经元突起长度,提取细胞的总RNA并纯化,设计合成G蛋白偶联受体(GPCR)基因的引物,采用Taqman荧光探针,实时定量PCR,检测离体培养海马神经元用药组和对照组细胞GPCR基因表达量的变化。结果用药组的神经元突起生长较长,GPCR的基因表达量显著高于对照组(P<0·01)。结论NRF使海马神经元生长并增加GPCR基因的表达,GPCR可引起神经细胞生长的生物效应。Objective To study the mechanism of the growth-promoting effects of Chinese herb "nerve regeneration faetor(NRY)" on the hippoeampal neurons. Methods Rat embryonic (17d) hippoeampl neurons were cultured in the presence or absence of NRF(1 μg/ml). The neurite length was measured, and total RNAs were extracted and purified from the control and NRF-treated neurons. The changes in gene expression of G protein-coupled reeeptor were detected with real time-PCR and specific primer and Taqrnan prob. Results The averaged neurite length of the neurons in the rats treated with NRF was significantly longer than that in the control (P〈0. 01). The gene expression of G protein-coupled receptor increased in the preseenee of NRF. Conclnsion NRF promotes neurite growth and upregulates gene expression of G protein-coupled receptor.
