摘 要:应用高速逆流色谱法(High-Speed Counter-current Chromatography,HSCCC),从丹参的乙醇粗提物中分离纯化脂溶性成分。试验研究建立了以石油醚-乙酸乙酯-甲醇-水为溶剂体系,并优化了HSCCC的操作参数及操作方法,快速有效地分离得到5个丹参酮类化合物。经HPLC方法测定,其中丹参酮ⅡA、隐丹参酮、丹参酮Ⅰ、二氢丹参酮Ⅰ纯度均大于98%。该方法操作简捷,快速高效、费用低廉,重现性好,为中药丹参的质量控制所需要的高纯度化学对照品,提供了一种优良的制备分离方法。Separation and preparation of tanshinones from Danshen (Salvia miltiorrhiza Bunge ) crude ethanol extract were studied by high-speed counter-current chromatography (HSCCC), The solvent system of petroleum ether-ethyl acetate-methanol-water (8:6:7:3) was selected and the optimum running parameters for apparatus was obtained through orthogonal experiment. Five tanshinone were separated and identified rapidly. Measured by HPLC, the purity of tanshinone Ⅱ A, cryptotanshinone, tanshinone Ⅰ , dihydrotanshinone Ⅰ were all over 98%. HSCCC is simple, economical, quick and with good repeatability for preparation of Danshen active components.
