摘 要:目的通过对照实验探讨自发性癫痫大鼠(SER)与野生型正常大鼠(WTC)海马Ⅰ型和Ⅲ型电压门控性钠通道α亚基mRNA与蛋白的表达情况。方法发现SER癫痫大发作后即刻提取海马组织总RNA,应用逆转录-聚合酶链反应(RT-PCR)表达α1与α3亚基mRNA;应用免疫荧光与免疫组化定位与表达α1亚基蛋白与α3亚基ⅢN蛋白。结果SER海马Ⅰ型与Ⅲ型钠通道α亚基mRNA表达高于WTC(P<0·01);SER海马Ⅰ型钠通道α亚基蛋白表达高于WTC(P<0·01);α3亚基ⅢN型蛋白在SER成鼠有表达,而WTC成鼠无表达。结论SER脑内海马Ⅰ型与Ⅲ型电压门控性钠通道α亚基表达上调,可能是神经元高兴奋性的原因或癫痫发作的继发表现。Aim To investigate mRNA and protein expressions of type Ⅰ and Ⅲ voltage-gated sodium channel α subunits in spontaneously epileptic rats (SER) and wild type control rats (WTC) hippocampus by control study. Methods Total RNA was extracted immediately from hippocampus after tonic convulsions of SER, and type Ⅰ and Ⅲ sodium channel α subunits mRNA expressions were detected by RT-PCR respectively. The location and expression of type Ⅰ and Ⅲ N sodium channel α subunits protein were detected by immunohistochemistry and immunofluoresence. Resuits Type Ⅰ and Ⅲ sodium channel α subunits mRNA of SER expressed significantly higher than those of WTC in hippocampus (P 〈 0.01 ). Type Ⅰ sodium channel protein of SER increased significantly in hippocampus ( P 〈 0. 01 ) , and Ⅲ N protein that should not have been expressed in adult WTC expressed in adult SER hippocampus. Conclusion The expression of type Ⅰ and Ⅲ sodium channel α subunits was regulated up in SER hippocampus, which may be the etiopathogenisis of neron hyperexcitability or succeeding appearance after seizure.
