摘  要:目的:通过转染野生型p53基因,并与5-Fu联合作用,观察其对两种鼻咽癌细胞生长的影响。方法:将克隆有野生型p53基因的pUHD10-3-p53质粒,用脂质体(Lipofectamine)介导分别转染人鼻咽癌CNE1和CNE2细胞,同时在培养液中加入5-Fu,用四甲基偶氮唑(MTT)法分析细胞生长情况。结果:人鼻咽癌CNE1和CNE2细胞分别加入含5-Fu的培养液后,第4天生长抑制率分别为49·46%、54·85%。CNE1和CNE2细胞分别转染wt-p53基因后,第4天生长抑制率分别为47·14%、45·90%。人鼻咽癌CNE1和CNE2细胞分别转染wt-p53基因后应用5-Fu处理,细胞的生长抑制率第4天分别可达到84·74%、85·82%,其生长作用均明显受到抑制。结论:野生型p53基因与5-Fu联合作用,对人鼻咽癌CNE1及CNE2细胞生长有明显的抑制效果。Objective: To investigate the effect of the combination of wild tape p53(wt-p53) gene transfected with 5-fluorouraeil(5-Fu) on the growth of nasopharyngeal carcinoma. Methods:The nasopharyngeal carcinoma lines CNE1 and CNE2 that were respectively transfeeted with plasmid pUHD10-3-p53 reeombined with wt-p53 gene by Lipofeetamine-mediate were cultured in medium containing 5-Fu. 3-(4,5-Dimethy-2- thiazolyl)-2,5-diphenyl-2H-tetrazolium-bromid(MTT) technique was used to analyse the growth of nasopharyngeal carcinoma lines CNE1 and CNE2. Result:The growth of nasopharyngeal carcinoma lines CNE1 and CNE2 were both inhibited after exposed to 5-Fu, and 49.46 %, 54.85 % of the cells were respectively inhibited on the fourth day. After transfected with wt-p53 gene,47. 14% and 45. 90% of the nasopharyngeal carcinoma lines CNE1 and CNE2 were inhibited on the fourth day. When the nasopharyngeal carcinoma lines CNE1 and CNE2 respectively transfeeted with wt-p53 gene had been exposed to 5-Fu for 4 days, 84. 74% and 85.82% of the cells were respectively inhibited. Conclusion.. The growth of the two nasopharyngeal carcinoma lines were obviously inhibited by the combination of wild tape p53(wt-p53) gene and 5- fluorouraeil(5-Fu).
