摘　　要：目的构成BCL-2基因表达的人胃黏膜上皮细胞系GES-1细胞模型。方法质粒pcDNA3-BCL-2经酶切鉴定后,以脂质体法转染人胃黏膜上皮细胞GES-1,经G418筛选获得细胞克隆,荧光免疫组化法鉴定BCL-2基因表达阳性细胞。结果经酶切鉴定为携带BCL-2基因的质粒pcDNA3-BCL-2,荧光免疫组化结果显示7个克隆中,均有不同程度的BCL-2基因表达。BCL-2主要表达在胞质及细胞核膜。结论GES-1细胞为BCL-2表达阳性的细胞,为进一步研究BCL-2基因与胃癌的关系提供了理想的细胞模型。Objective To establish a Human Gastric Epithelial cell strain GES-lstably transfected by BCL-2 gene. Methods After the pc^DNA3-BCL-2 was identified by restriction endocleavase digestion, lipofectamine method was adopted to transfect pc^DNA3-BCL-2 into human gastric epithelial cell line GES-1. The geneticin 418 （ G418 ） was used to select positive clones. F1TC labeled immunohistological staining was used to detect BCL-2 protein positively expressed cells. Results Identified by restriction endocleavase digestion, it was the plasmid carrying BCL-2 gene. BCL-2 protein positive expression in 7 transfected cell clones were identified by FITC labeled immunohistological staining. BCL-2 protein was expressed in cytoplasm and nucleomembrane. Conclusion These cells are BCL-2 protein positively expressed cells. The establishment of this cell model can be used to further study of the role of BCL-2 gene in gastric carcinoma.
