摘  要:目的探讨高脂饮食诱导下肥胖易感(obesityprone,OP)与肥胖抗性(obesityresistant,OR)大鼠白色脂肪组织(whiteadiposetissue,WAT)中脂肪酸合成酶(fattyacidsynthase,FAS)的表达差异。方法72只SD大鼠,随机选取12只为标准对照组,基础饲料饲养7周,余60只基础饲料饲养1周后,按体重增值分为OP组、OR组和高脂组,分组后改用高脂饲料继续饲养6周。观察各大鼠体重变化、Lee’s指数、脂肪湿重等;用RTPCR技术分析WAT中FAS基因mRNA表达水平,Westernblotting检测WAT中FAS的蛋白表达差异。结果OP组体重增值、Lee’s指数及脂肪湿重均高于OR组(P<0.05);OP组FAS基因mRNA表达水平高于OR组(P<0.05);OP组FAS蛋白表达显著高于OR组(P<0.001)。结论OP与OR大鼠的白色脂肪组织中存在FAS基因表达差异,其差异与大鼠发生肥胖的易感程度有关。Objectives To explore the different expression of fatty acid synthase (FAS) in white adipose tissue (WAT) of obesity-prone (OP) and obesity-reslstant (OR) rats on a high-fat diet. Methods After one week of free access to a standard diet, 12 rats were selected randomly as standard control group and were fed on standard diet, the 60 remaining rats were separated on the basis of one week body weight percentage gained to OP or OR group, and were being fed on high-fat diet for another 6 weeks, The body weight, Lee' s index, the wet weight of epitesticle, retroperitoneal and subcutaneous fat pad were measured after sacrificing the rat. The expression of FAS mKNA in WAT was detected by RT-PCR technique and the expression of FAS protein was determined by Western blotting. Results In OP rats, the speed of body weight gain, the Lee' s index and the wet weight of fat pad at cpitesticle, retroperitoneal and subcutaneous sites were higher than those in OR rats (P 〈 0.05) . There were higher levels of FAS mRNA expression in OP rats than in OR rats. The OP rats showed higher expression of FAS protein in WAT than OR rats. Conclusions Different expression of FAS in WAT existing between high-fat induced obesity-prone and obesity-rcsistant rats suggest that FAS may be involved in tho susceptibility to obesity.
