肿瘤坏死因子-α基因与精神分裂症传递不平衡的研究

摘　　要：目的 探讨肿瘤坏死因子-α基因（TNF-α）-C863A和-G308A多态性位点在精神分裂症发病机理中的作用。方法 收集172个来自广东潮汕地区的精神分裂症核心家系，采用聚合酶链式反应-限制性片段长度多态性方法，检测所有研究对象的肿瘤坏死因子-α基因-C863A和-G308A多态性位点的基因型，并进行单体型相对风险和传递不平衡检验。结果 单体型相对风险分析：-C863A位点x^2=0．05，P〉0．05，-G308A位点x^2=0．87，P〉0．05。单位点传递不平衡检验：-C863A位点x^2=0．19，P〉0．05，-G308A位点x^2=0．80，P〉0．05；多位点单体型分析结果表明，由这两个位点组成的单体型不存在过度传递。在106家以阳性症状为主的精神分裂症家系中，有一条单体型，C863-A308，其x^2=4．00，P〈0．05，表明该单体型在以阳性症状为主的精神分裂症家系中过度传递。结论 在广东潮汕人群中，肿瘤坏死因子α基因可能在阳性症状为主的精神分裂症患者发病机制中起一定作用。Objective To explore the relationship between the polymorphism loci -C863A and -G308A in the tumor necrosis factorctgene （TNF-α） and schizophrenia. Methods Using PCR-restrietion fragment length method, we genotpyed the polymorphism loci -C863A and -G308A in TNF-α gene in 172 schizophrenic trio families collected from Chaoshan region. The data were analyzed with Haplotype relative risk and Transmission/disequilibrium test. Results The Haplotype relative risk revealed that the polymorphism loci -C863A （x^2 = 0.0502, P 〉 0.05 ） and -G308 A （ x^2 = 0.80 , P 〉 0.05 ） no significantly preferential of any allelie frequency for the two polymorphism loci in TNF-α gene. The one-maker Transmission Disequilibrium Test （TDT） suggested that the two polymorphism loci -C863A （ x^2 = 0.19, P 〉 0.05 ） and -G308A （ x^2= 0.80 , P 〉 0.05 ） in TNF-α gene had not biased transmission. The multi-maker haplotypes analysis results showed there were no haplotypes existed over transmission; In 106 schizophrenia parent-offspring trios that the positive symptomatic as major features analysis suggested there was a haplotype C863-A308, x^2 = 4.00, P 〈 0.05, which existed over transmission in the positive symptomat- ic as major features patients. Conclusion The results suggested that TNF-α gene may be play a role in the pathogenesis of schizophrenia that the positive symptomatic as major features in the Chaoshan population.
