摘 要:目的研究RASSF1A基因甲基化状态及其基因表达异常与口腔癌前病变及鳞癌发生发展的关系。方法采用甲基化特异性PCR(MSPCR)技术检测了10例正常口腔粘膜、8例上皮单纯增生、20例上皮异常增生、32例鳞癌组织中RASSF1A基因甲基化状况;运用逆转录-聚合酶链反应(RT-PCR)研究口腔粘膜癌前病变及鳞癌组织中RASSF1A基因mRNA的表达水平。结果10例正常口腔粘膜无甲基化,且RASSF1A基因mRNA呈100%表达;8例上皮单纯增生中,有1例甲基化,1例RASSF1AmRNA表达下降;20例上皮异常增生中,有3例甲基化,5例RASSF1AmRNA表达下降;32例鳞癌组织中,有13例甲基化,占40.63%,15例RASSF1AmRNA表达下降或无表达,占46.88%。结论RASSF1A基因甲基化是口腔癌癌变的早期事件,与口腔鳞癌的发生发展有关。RASSF1A基因甲基化与RASSF1A基因转录抑制高度相关。Objective To analyse the relationship between methylation status of the 5 CpG island and mRNA expression of the RASSF1A gene in normal oral mucosa (NOM), oral premalignant lesions and oral squamons cell carcinomas (OSCC). Methods Metylation status of RASSF1A gene was determined by methylationspecific PCR, and RASSF1A mRNA expression was evaluated by RT- PCR in a series of 70 specimens, including 10 NOM, 8 epithelial hyperplasia, 20 premalignant lesions and 32 OSCC. Results Ten specimens of NOM displayed no methylation and showed normal expression of the RASSF1A mRNA. One of 8 epitbelial hyperplasia was methylation of RASSF1A exon 1, 1 of 8 epithelial hyperplasia showed decrease of RASSF1A mRNA expression. Three of 20 oral premalignant lesions were methylation of RASSF1A exon 1, 5 of 20 oral premalignant lesions showed decrease of RASSF1A mRNA expression; 13 of 32 OSCC were methylation of RASSF1A exon 1, 15 of 32 OSCC showed decrease of loss of RASSF1A mRNA expression. Conclnsion Methylation of RASSF1A gene is an early event in the malignant progression of OSCC. Methylation is highly correlated with inhibition of RASSF1A gene transcription.
