摘 要:目的:探讨总抗氧化能力、丙二醛、脂质过氧化物对诊断超声照射后大鼠生殖细胞凋亡的影响。方法:SD雄性健康大鼠32只,用诊断超声照射不同时间,24 h后取大鼠睾丸组织,用生物素-dUTP酶标亲和素法(TUNEL法)检测细胞凋亡率(AI),化学比色法测定总抗氧化能力(T-AOC)、丙二醛(MDA)、脂质过氧化物(LPO)。结果:诊断超声照射10 min组,睾丸组织中T-AOC(IU/g)为36.4±4.6、MDA(μmol/g)为1.62±0.53、LPO(μmol/g)为1.83±0.35、睾丸生殖细胞凋亡(%)为6.6±0.35,分别与对照组(34.5±3.5,1.25±0.45,1.78±0.27,5.8±0.21)比较,差异无统计学意义(均为P>0.05),而超声照射20 min组T-AOC(IU/g)为18.3±2.8、MDA(μmol/g)为8.56±0.68、LPO(μmol/g)为7.36±0.28、细胞凋亡(%)为16.3±3.67;30 min组的睾丸组织中T-AOC(IU/g)为16.2±1.7、MDA(μmol/g)为8.13±0.75、LPO(μmol/g)为7.28±0.37、细胞凋亡(%)为18.5±2.89。超声照射20 min组及30 min组T-AOC显著降低,MDA、LPO含量升高,睾丸生殖细胞凋亡增加,与对照组比较,差异有统计学意义(均为P<0.01)。结论:诊断超声照射20 min以上,可导致大鼠睾丸生殖细胞凋亡增加,且与睾丸组织中总抗氧化能力下降、丙二醛及脂质过氧化物升高密切相关。Objective: To study the effect of oxidation/antioxygen system on the germ cell apoptosis after diagnostic ultrasound radiation. Methods: Thirty-two immature male healthy rats (Sprague- Dawley) were administered diagnostic ultrasound radiation for different time. The levels of total antioxidant capacity (T-AOC), malondialolehyde (MDA) and lipidperoxide (LPO) in testis were evaluated by spectrophotometric method. Germ cell apoptosis rates were detected by TdT-mediated dUTP-x nick end labeling (TUNEL). Results: The level of T-AOC (IU/g), MDA (mol/g), LPO (μmol/g) and the apoptosis rate (%) was 36.4±4.6, 1.62±0. 53, 1.83±0.35 and 6.6±0. 35 respectively in 10-min-irradiation group; the indices were 18.3±2.8, 8. 56±0. 68, 7. 36±0.28 and 16.3±3.67 respectively in 20-min-irradiation group; 16.2± 1.7, 8.13±0.75, 7.28± 0.37 and 18.5±2.89 respectively in 30-min-group; and 34.5±3.5, 1.25±0. 45, 1.78±0. 27, and 5.8 ± 0. 21, in the control group. There were significant differences between each of the groups for irradiating for at least 20 minutes and the control group in these indices (all P〈0.01). While no significant difference between the 10-min-irradiation group and control group was found (all P〉0.05). Conclusion. If the time of diagnostic ultrasound radiation is beyond 20 minutes, testicle apoptosis rate is increasing along with the descending antioxygen capability, MDA and LPO.
