RI与TACE联合栓塞对大鼠Walker-256肝移植模型肿瘤细胞增殖和凋亡的影响

摘　　要：目的研究核糖核酸酶抑制因子(RI)与TACE联合栓塞Walker-256肝移植模型对肿瘤细胞增殖和凋亡的影响。方法将60只Walker-256荷瘤大鼠于移植后第14天随机分成肝动脉灌注化疗栓塞组(TACE组)、RI与TACE联合栓塞组和对照组,每组20只。各组分别经肝动脉注入:超液态碘油0.5 ml加5-Fu 20 mg/kg;RI 200单位加5-Fu 20mg/kg加超液态碘油0.5 ml;生理盐水0.5 ml。于术后第7天用TUNEL法检测肿瘤细胞的凋亡率,用免疫组化法检测肿瘤细胞PCNA增殖率、平均染色强度。结果PCNA增殖率和平均染色强度TACE组为44.98±7.92,168741.41±25410.37;联合栓塞组为45.02±8.42,168539.41±19675.98;对照组为35.43±6.75,137313.01±17380.59。TACE组、联合栓塞组PCNA增殖率和染色强度显著高于对照组(P<0.01),而联合栓塞组与TACE组差异无统计学意义(P>0.05)。肿瘤细胞凋亡率TACE组为16.88±2.48;联合栓塞组为23.50±5.33;对照组为11.70±2.76,TACE组、联合栓塞组显著高于对照组(P<0.01),联合栓塞组显著高于TACE组(P<0.01)。结论RI与TACE联合栓塞与TACE相比可显著提高肿瘤细胞的凋亡率。Objective To investigation the expression of pmliferaton and apoptosis after combination embolization with fibonudease inhibitor plus TACE in the rat walker-256 bepatoma nmdel.Methods The 60 rats of walker-256 bepatoma model were divided into 3 groups at random after the 14th day of the tumors were implanted. Hepatic artery embolizationwith 5-Fu 20 mg/kg and lipiodol 0.5 ml in TACE group,RI 200 U and TACE in the combination embolization group,saline solution 0.5 ml in control group. On day 7 after embolization, file apoptosis rate of tumors tissue was assessed with the method of TUNEL, PCNA proliferation rate, staining intensity assessed applying to immunohistochemical methods.Results The PCNA proliferation rate and average staining intensity were 44. 98 ± 7.92 vs 168741.41 ± 25410.37 in TACE group; 45.02 ± 8.42 vs 168539.41 ± 19675.98 in combination embolization group; 35.43 ±6.75 vs 137313.01 ± 17380.59 in control gronp.The rate of proliferaton was signifinant enhance in TACE group and combination embolization group compare with control group（ P 〈 0.01） ,but no difference from between TACE group and comgination embolization group （P 〉 0.05 ）. The rate of apoptosis was 16.88 ± 2.48,23.50 ± 5.33 and 11.70 ± 2.76 in TACE group, combination embolization group and control group, respectively. The apoptosis rate was signifinant enhance in TACE group and combination embolization group compare with control group（ P 〈 0.01 ）, there were also signifinant different between TACE group and combination embolization group（ P 〈 0.01）. Conclusion combination embollzation might significant enhance the rate of apoptosis on tumor cell compare with TACE.
