摘  要:目的研究氦氖激光治疗阿尔茨海默病(AD)模型大鼠后脑组织微管相关蛋白2(MAP2)及凋亡相关基因bc l-2表达的变化。方法SD大鼠40只,随机分为假损伤组、AD模型组、氦氖激光非穴位治疗组和氦氖激光穴位治疗组4组,每组10只。采用大鼠脑组织立体定位微量注射技术,用鹅膏蕈氨酸(IBO)损毁大鼠双侧迈纳特基底核。术后用氦氖激光照射AD大鼠“百会”、“大椎”穴进行治疗,用非穴位组作为对照。造模23 d后,免疫组织化学染色检测脑组织微管相关蛋白MAP2及凋亡相关基因bc l-2的表达。结果氦氖激光穴位治疗组脑组织MAP2阳性反应光密度值较AD模型组和非穴位治疗组明显增高(P<0.05),与假损伤组比较无显著性差异(P>0.05);氦氖激光穴位治疗组bc l-2表达明显高于假损伤组和AD模型组(P<0.05),与非穴位治疗组无显著性差异(P>0.05)。结论氦氖激光穴位照射可促进树突的生长和受损结构的修复,并通过上调抗凋亡基因bc l-2表达而实现对AD鼠的改善。Objective To study the effect of He-Ne Laser on the expression of mierotubule-associated protein-2 (MAP2) and bcl-2 in brain tissue of model rats with Alzheimer disease (AD). Methods Fourty SD rats were randomly divided into four groups, sham operated group, AD model group ,the group treated with He-Ne Laser not at "Baihui" and "DaZhui" points, the group treated with He-Ne Laser at "Baihui" and "Dazhui" points synchronously, each group contained 10 rats. The AD rat model was established by injecting IBO to rats brain to damage bilateral nbM in rats brain. 23 days after the AD rat model were established, all of the rats were isolated for MAP2 and bcl-2 by imraunohistochemistry stain. Then the gray scale value of MAP2 positive cell and the number of bcl-2 positive neuron were detected. Results Compared with AD model group and the group treated with He-Ne Laser not at "Baihui" and "Dazhui" points,the average optical density(OD) of MAP2 positive cells of the group treated with He-Ne Laser at "Baihui" and "Dazhui" was increased (P 〈 0. 05 ), but there wasn't distinct difference with sham operated group ( P 〉 0.05 ) ; Compared with sham operated group and AD model group, the number of bcl-2 positive neuron in the group treated with He-Ne Laser at "Baihui" and "Dazhui" points increased markedly ( P 〈0. 05 ), but there wash' t distinct difference with the group treated with He-Ne Laser not at "Baihui" and "Dazhui" points ( P 〉 0. 05). Conclusion He-Ne laser irradiation may improve dendritic cell growth and injured structure's resume, and increase the expression of anti-apoptosls gene bcl-2.
