摘 要:目的探讨肿瘤细胞裂解物致敏的树突状细胞(DC)作为佐剂对肝癌HepG2细胞系的杀伤作用。方法反复冻融法裂解培养的肝癌HepG2细胞提取细胞抗原,然后致敏DC,观察后者对自身T淋巴细胞增殖的影响以及T细胞对HepG2细胞的杀伤作用。结果肝癌HepG2细胞裂解物致敏的DC能显著促进T细胞的增殖,后者对HepG2细胞有明显的杀伤效应。结论肿瘤细胞裂解物提取方法简单,其作为细胞抗原致敏DC制备的肿瘤疫苗能保证抗肿瘤免疫反应的全面性和有效性,并降低可能出现的肿瘤免疫耐受,将有很好的临床应用前景。Objective: To detect the anti - hepatic cancer ability of whole cell lysates pulsed dendritic cell ( DC) acted as an adjunvant. Methods: Whole cell lysates of HepG2 cells were extracted with freeze thawing method, then the monocyte - derived DCs were pulsed with this cellular antigen. Subsequently, The capability of antigen pulsed DCs to promote T lymphocyte proliferation and the ability of T cells to kill HepG2 cells were detected by using 3H - TdR incorporation and LDH release methods, respectively. Results: The whole cell lysates of HepG2 cells pulsed DCs could significantly stimulated T cells proliferation, and the cytotoxieity abilities of primed T cells to kill HepG2 cells were also enhanced. Conclusion: Tumor cell lysates acted as the cellular antigen to pulse DCs could improve the efficacy of anti - cancer immune response and decrease the anti - cancer immune tolerance, which provided us an experimental evidence for cancer immunotherapy.
