摘 要:目的探讨细胞凋亡及其效应分子Caspase-3在梗阻性肾病中的作用及其机制。方法应用免疫组化方法和原位细胞凋亡检测法分别检测43例梗阻肾组织和10例正常肾组织中Caspase-3表达和细胞凋亡指数,并分析梗阻肾Caspase-3表达与肾积水程度及肾纤维化程度之间关系。结果梗阻肾的肾小管和间质中细胞凋亡指数和Caspase-3表达均显著高于正常肾组织(P<0.05),而且Caspase-3表达与细胞凋亡指数密切正相关(P<0.01),与肾积水程度也密切正相关(P<0.05);梗阻肾间质Caspase-3表达与肾纤维化程度密切相关(P<0.05)。结论Caspase-3活性增强导致细胞凋亡增多在梗阻肾病理生理过程中起着重要作用。Objective To investigate the roles and mechanisms of apoptosis and its effector molecule Caspase-3 in the pathogenesis of chronic obstructive kidney. Methods Caspase-3 expressions in 43 obstructive renal tissues and 10 normal renal tissues were examined by immunohistochemistry, and apoptosis was examined by TUNEL methods. The relationships between Caspase-3 expression of obstructive renal tissues and degree of hydronephrosis and renal fibrosis were evaluated. Results Both Caspase-3 expressions and apoptosis indexes in tubular and interstitial cells of obstructive kidneys were significantly higher than those in normal kidneys (P 〈 0.05). Caspase-3 expressions in obstructive kidneys were in close association with apoptosis and hydronephrosis (P〈0.05). Caspase-3 expressions in interstitial cells of obstructive kidneys were closely related to the degree of renal fibrosis (P〈0.05). Conclusion Increased Caspase-3 activity plays an important role in renal cell apoptosis associated with urinary obstruction.
