摘 要:目的研究葡萄籽提取物(GSE)对四氯化碳(CCl4)诱导肝损伤的肝功能保护作用。方法建立CCl4诱导肝损伤的大鼠模型,测定正常对照组、四氯化碳诱导组(CCl4组)、GSE低剂量组(20mg/kg·d)、GSE中剂量组(40mg/kg·d)和GSE高剂量组(120mg/kg·d)共5个组血清中碱性磷酸酶(ALP)、天门冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、γ-谷氨酰转移酶(γ-GT)、谷氨酸脱氢酶(GDH)、异柠檬酸脱氢酶(ICDH)和亮氨酸转肽酶(LAP)水平的改变,称量肝重和体重,比较各组大鼠肝体比值、甘油三酯(TG)及肝组织病变积分的改变。结果CCl4组的血清ALT、AST、GDH和ICDH上升,明显高于其它组(P<0.01、P<0.01、P<0.01和P<0.01),该组血清LAP和!-GT也高于正常对照组(P<0.05和P<0.05)、血清ALP明显高于正常对照组和高剂量组(P<0.01和P<0.01);低剂量组和中剂量组的血清ICDH高于正常对照组(P<0.01和P<0.01)、高剂量组则与对照组无差异(P>0.05);低剂量组和中剂量组的血清ALP高于正常对照组(P<0.01和P<0.01)、高剂量组则与对照组无差异(P>0.05)。四个组的肝体比值未见明显差异(P>0.05)。CCl4组的肝脏中TG值高于正常对照组和中剂量组(P<0.05和P<0.05)。GSE高、中、低剂量组肝细胞病变积分均低于CCl4组(P<0.05、P<0.05和P<0.05)。结论葡萄籽提取物可降低肝细胞受损引起的各种酶的血清浓度上升,并且明显改善肝脏病理改变。Objective To study the protective effects of grape seed extractive (GSE) on carbon tetrachloride (CCl4) induced hepatic injury. Methods CCl4 induced hepatic injured rats were divided into CCl4 group (untreated) and 4 treatment groups treated with 20 mg/kg·d low dose group (LDG), 40 mg/kg·d medium dose group (MDG) or 120 mg/kg-d high dose group (HDG). In these five groups the serum alkaline phosphatase (ALP), aspartic transaminase( AST ), alanine aminotransferase ( ALT ),γ-giutamyltransferase (γ-GT ), glutamate dohydrogenase (GDH), isocitrate dehydrogenase (ICDH)and amidocaproic acid transpeptidase (LAP)were measured. The liver weight to body weight ratio, serum triglycerides (TGs), and the pathological changes were also studied. Results Serum levels of ALT, AST, GDH and ICDH in control group were increased significantly and were higher than the other four treatment groups(P〈0.01). The serum levels of LAP and T-GT were higher than normal group(P〈0.05). The serum level of ALP in CCl4 group was also significantly higher than normal group (P〈0.01) and HDG group(P〈0.01 ). Serum level of ICDH in LDG and MDG were higher than normal group (P〈0.01)with no difference between HDG and normal group (P 〉 0.05). Serum ALP in LDG and MDG were higher than normal group (P〈0.01 and P〈0.01) but showed no difference between HDG and normal greup(P 〉0.05). No significant differences were observed on liver weight and body weight ratio among the five groups (P 〉0.05). The serum trigiycefides level of the CCl4 group was significantly higher than normal group and MDG(P〈0.05). Pathological change of threc GSE groups were lower than CCl4 group(P〈0.05,P〈0.05 and P〈 0.05). Conclusion Grape seed extractive can lower the levels of serum triglycerides and MDA and reduce the pathological changes, thus protect the liver from CCl4 induced hepatic injury.
