西比灵干预对沙鼠脑内COX-2及PAI-1表达影响的研究

摘  要:【目的】研究西比灵对沙鼠脑缺血后脑内环氧合酶-2(COX-2)和1型纤溶酶原激活物抑制剂(PAI-1)蛋白表达变化的影响。【方法】制作沙鼠脑缺血再灌注模型;实验前给沙鼠喂食西比灵;分别在缺血再灌注(IR)d1、d3、d7用免疫组织化学方法检测脑内COX-2和PAI-1蛋白的表达,并与其他各组比较。【结果】沙鼠脑缺血再灌注后COX-2和PAI-1均明显表达,COX-2随再灌注后时间的延长而递减,而PAI-1却递增(P<0.01)。西比灵干预后COX-2和PAI-1表达趋势同缺血再灌组,COX-2阳性率低于缺血再灌注组;而PAI-1阳性率高于缺血再灌组;两者均有显著性差异(P<0.01)。【结论】沙鼠脑缺血后可诱导表达COX-2和PAI-1蛋白,西比灵可下调COX-2而上调PAI-1蛋白表达,有利于抵御脑缺血再灌注后脑损伤。[Objective]To study the effects of flunarizine on the expressions of COX-2 and PAF1 proteins in brain tissue following transient cerebral ischemic reperfusion in gerbils. [Methods]A cerebral transient isehemic-reperfusion model in gerbils was established by clamping both common carotid arteries; One group of gerbils were fed with flunarizine before experiment. The expressions of COX-2 and PAI-1 protein in brain tissue were detected by immunohistochemistry technique at the 1st, 3rd and 7th day after ischemic reperfusion. [Results] COX-2 and PAI-1 proteins were expressed in neurons and gliocytes of the brain tissues in 30 gerbils. Compared with normal control group, the expressions of COX-2 and PAI-1 proteins in neurons and gliocytes of ischemic gerbils were evidently increased at the 1st , 3rd and 7th day( P〈0. 01). Compared with cerebral ischemia group, the expressions of COX-2 protein in neurons and gliocytes of gerbils in flunarizine treatment group were evidently decreased at the 1st , 3rd and 7th day, while PAI-1 protein expression was obviously increased ( P 〈0.01). [Conclusion]Expressions of COX-2 and PAI-1 proteins in neurons and gliocytes of gerbils are evidently increased following cerebral ischemic reperfusion',the expression of COX-2 protein in neurons and gliocytes of gerbils in flunarizine treatment group is evidently decreased while PAI-1 protein is evidently increased. Flunarizine can down-regulate the expression of COX-2 protein and up-regulate the expression of PAI- 1 protein.
