摘 要:目的研究CD44和透明质酸(HA)与大肠癌浸润、转移的关系及二者表达的相关性。方法收集33例大肠癌患者肿瘤组织和远癌切端正常组织标本,采用免疫组织化学方法检测细胞膜上CD44蛋白及细胞外基质中HA的表达,同时用逆转录-聚合酶链反应(RT-PCR)检测CD44的mRNA表达。结果大肠癌组织中CD44的mRNA转录水平和细胞膜上蛋白表达水平、HA在肿瘤基质的蛋白表达水平均高于远癌切端正常组织;CD44的mRNA转录水平和细胞膜上蛋白表达水平与大肠癌的淋巴结转移、Dukes'分期密切相关;HA在大肠癌细胞外基质(extra-cellularmatrix,ECM)中的表达水平与淋巴结转移、Dukes'分期显著相关。结论大肠癌中HA与其受体CD44结合后,促进了肿瘤的浸润和转移;同时检测CD44与HA可预测大肠癌转移潜能,评估患者的预后。[Objective] To individually research the correlations between CD44, hyaluronic acid (HA) and the infiltration and metastasis of colorectal carcinoma (CRC), and to study the correlation of expressions of the two factors. [Methods] The expressions of CD44 protein on the cell membrane and HA on extracellular matrix (ECM) of tumor and corresponding normal control tissues collected from 33 patients with CRC who underwent surgical procedure were assayed by immunohistochemistry. Meanwhile the gene expression levels of CD44 were demonstrated by reverse transcription polymerase chain reaction (RT-PCR). [Results] The mRNA transcription and protein expression levels of CD44 on cell membrane and HA in ECM of CRC were significantly higher than those in normal control tissues. There were significantly correlations between the mRNA transcription and protein expression levels of CD44 on CRC cell surface and lymphatic metastasis as well as Dukes' stage. The expression of HA in the ECM of CRC was correlated with lymphatic metastasis and Dukes' stage. [Conclusions] In CRC, HA combined with its receptor CD44 promotes infiltration and metastasis of the tumor cell. To assay HA and CD44 together might be a factor to evaluate the metastasis potency and helpful for prognostic evaluation.
