摘  要:目的:探讨缺氧对乳腺癌细胞(MDA-MB-435细胞)肌动蛋白细胞骨架重构和细胞形态改变与RhoA-ROCK信号通路的关系。方法:对体外培养的MDA-MB-435细胞进行缺氧16h处理,或在缺氧前用ROCK的特异性抑制剂Y-2763(210μmol/L)预处理细胞1h,再进行缺氧处理。用共聚焦激光扫描显微镜研究缺氧或使用Y-27632对MDA-MB-435细胞肌动蛋白细胞骨架重构的影响和细胞形态的改变。结果:乳腺癌细胞经16h缺氧处理,肌动蛋白细胞骨架发生解聚,应力纤维的排列和分布以及细胞形态明显改变;事先使用Y-27632再进行缺氧处理,可防止缺氧所致乳腺癌细胞内肌动蛋白细胞骨架解聚,同时应力纤维在细胞内排列和分布恢复正常。结论:RhoA-ROCK信号通路可能参与缺氧所致乳腺癌细胞内肌动蛋白细胞骨架的重构和细胞形态的改变。Objective:To investigate the role of RhoA-ROCK signal pathway in the reorganization of actin cytoskeleton and the ehanges of morphology induced by hypoxia in breast cancer cells (MDA-MB-435 cells). Methods:During the experiments the breast cancer cells were exposed to hypoxia for 16 h or pre-incubated with a special inhibitor of ROCK, Y-27632( 10 umol/L) for lh before hypoxia in vitro. The reorganization of cellular aetin cytoskeleton and the changes of cellular morphology induced by hypoxia or pretreatment of Y-27632 were examined by confocal laser scanning microscopy. Results:Hypoxia induced the depolymerization of aetin eytoskeleton, and the changes of arrangement and distribution of stress fihers, and the changes of morphology were observed obviously in breast cancer cells. Pretreatment with Y-27632 (10 umol/L) before hypoxia could prevent the depolymerization of actin eytoskeleton and the changes of arrangement and distribution of stress fihers induced by hypoxia in hreast cancer cells. Condusion:RhoA- ROCK signal pathway may mediate the reorganization of actin cytoskeleton and morphological changes in breast cancer cells during hypoxia.
