春性甘蓝型油菜小孢子培养技术研究

摘　　要：对一批春性甘蓝型油菜(Brassica napus)杂种F1及其亲本进行小孢子培养的结果表明,不同组合(基因型)出胚率差异比较大,2006年22份材料(基因型)中有21份出胚,其中40.91%的材料每蕾出胚数在5个以上,31.82%的材料每蕾出胚数在10个以上;杂交组合的出胚率大部分高于其亲本,主枝的产胚率绝大部分都高于其分枝;秋水仙碱不同处理中以0.6 g/L浓度处理5.0 h和3.4 g/L浓度处理1.5 h,植株成活率和加倍率最高;经秋水仙碱加倍后可育的植株大部分花粉活力较高,结实指数较高;而半可育的植株花粉活力低,并有50%的植株结实指数低于1。Studies on microspore culture of a set of F1 hybrids of spring-planted Brassica napus and their parents. The results indicated that the difference of ratio of embryos among different genotypes was much great. In all of the 22 donor materials, there were 21 of them being obtained embryos in 2006, average embryos per bud of 40.91% and 31.82% of which were over 5 and 10, respectively. The ratios of embryos of most F1 hybrids and the terminal raceme were higher than their parentsr and primary branchesp, respectively . In the treatments of 0.6 g/L colchicines for 5.0 h and 3.4 g/L col- ehicines for 1.5h, the effieiencies of survival and doubling were the highest. There were fertile plants and semi-fertile plants after doubling, the pollen vigor of the former was higher than the latter＇s ,furthermore, the selfing seed-set index of the former was high, too. And the selling seed-set index of 50% semi-fertile plants was lower than 1.
