摘　　要：目的探讨子宫内膜癌组织中PTEN mRNA和PTEN蛋白的表达及其临床意义。方法应用RT-PCR方法检测50例子宫内膜癌和15例正常子宫内膜组织中PTEN基因外显子1-8和5-9片段mRNA的表达;应用免疫组化技术检测PTEN蛋白的表达。结果(1)子宫内膜癌组织中,PTEN mRNA的表达缺失率在外显子1-8和5-9分别为48.00%和38.00%,正常子宫内膜组织PTEN mRNA全部呈阳性表达。两者在两组中表达缺失率的差异均有显著性(P<0.05);PTEN mRNA的表达缺失率在不同组织学类型、FIGO分期和组织学分级有随肿瘤恶性程度的增高而增高的趋势。(2)子宫内膜癌组织中,PTEN蛋白表达缺失率为60.00%,高于正常子宫内膜组织(0,P<0.01);PTEN蛋白表达缺失率在G3级组织和特殊类型癌中分别高于G1～G2级组织和内膜样腺癌(P<0.05)。(3)子宫内膜癌组织中,PTEN mRNA表达缺失和PTEN蛋白表达缺失呈正相关(P<0.01)。结论PTEN mRNA和蛋白表达缺失在子宫内膜癌的发生发展中可起重要作用。Objective To study the expression of PTEN mRNA and PTEN protein in endometrial carcinoma. Methods Expression of PTEN mRNA was examined by RT-PCR analysis. Expression of PTEN protein was examined by immunohistochemistry. Results The deletion of PTEN mRNA expression in endometrial carcinoma（exon 1-8 48. 00%, exon 5-9 38. 00%） was higher than that of normal tissue（P〈0. 05）. The PTEN mRNA expression deletion was irrelevant to histological types,FIGO stages and histological grades in endometrial carcinoma. The PTEN protein expression deletion in endometrial carcinoma（60. 00%） was higher than that of normal tissue（P〈0. 01）. The PTEN protein expression deletion was positively correlated to histological types and histological grades in endometrial carcinoma. Conclusion The PTEN mRNA and PTEN protein expression deletion may play an important role in the genesis and development of endometrial carcinoma.
