摘 要:目的探讨肺腺癌中环氧化酶2(COX-2)和缺氧诱导因子1α(HIF-1α)表达的相关性。方法通过免疫组化染色检测106例肺腺癌手术标本中COX-2和HIF-1α的表达,应用不同浓度的选择性COX-2抑制剂NS398对在正常和氯化钴模拟缺氧条件下培养的肺腺癌A549细胞进行处理,Westernblot法分别检测各组HIF-1α的表达。结果106例标本中COX-2强阳性表达率41.5%(44/106),弱阳性表达率20.8%(22/106),阴性表达率7.7%(40/106);HIF-1α强阳性表达率34.0%(36/106),弱阳性表达率4.0%(36/106),阴性表达率2.1%(34/106)。COX-2和HIF-1α的表达呈正相关(P<0.01)。常氧条件下HIF-1α表达相对值0.185±0.057,但缺氧条件下则升高至1.011±0.131,而NS-398则可以明显逆转其趋势(P<0.05)。结论COX-2与HIF-1α在肺腺癌中表达正相关,NS-398不仅能够抑制常氧条件下HIF-1α的表达,而且能够剂量依赖性的抑制低氧条件下HIF-1α表达上调。Objective To investigate the relationship of COX-2 and HIF-1α in human lung adenocarcinoma and the possible antineoplastic mechanism of selective COX-2 inhibitor. Methods Immunohistochemical method was employed to detect the expression of COX-2 and HIF-1α in tumor tissue of 106 patients. Western blot was employed to evaluate the effects of variant concentration of NS-398 on the expression of HIF-1α in cohalt chloride-stimulated A549 cell. Results Among 106 tumor tissues, the expression of COX-2 was strongly positive stained in 41.5% ,positive stained in 20. 8%, negative stained in 37.7%. Meanwhile, the expression of HIF-1α was strongly positive stained in 34.0% ,positive stained in 34. 0% ,negative in 32. 1%. The expression of COX-2 was positively correlated with HIF-1α(r= 0. 440, P〈0. 01). The expression of HIF-1α increased greatly from 0. 185 ± 0. 057 (no cobalt chloride-stimulated) to 1. 011 ±0. 131 (cobalt chloride-stimulated), and NS-398 could inhibit the expression of HIF-1α in either condition(P〈0. 05). Conclusion High expression of COX-2 is correlated with HIF-1α. NS-398 could inhibit the expression of HIF-1α in a dose-dependent manner in cobalt chloride-stimulated A549 cells.
