摘 要:目的研究糖尿病大鼠视网膜中缺氧诱导因子-1α(HIF-1α)的表达与超微结构变化。方法将132只SD大鼠随机分为对照组与糖尿病视网膜病变(DR)组,DR组采用链脲佐菌素(STZ)一次性腹腔注射建立糖尿病模型。应用免疫组织化学SP法和RT-PCR技术分别于1、3、6个月检测视网膜中HIF-1α蛋白及其mRNA的表达,透射电镜观察视网膜的超微结构。结果免疫组织化学与RT-PCR检测结果均显示,HIF-1α蛋白及其mRNA在对照组视网膜中不表达,在DR组中呈进行性表达增强(P<0.05);透射电镜观察正常对照组大鼠视网膜血管未见异常,DR组随着病程发展逐渐出现视网膜血管内皮细胞肿胀,基底膜厚薄不均。结论DR中HIF-1α的异常表达和超微结构改变与视网膜缺氧关系密切,两者与DR的发生发展密切相关。Objective The pathogenesis of diabetic retinopathy is still insufficiently understood, although some reports have implicated the role of the chronic isehemia. This research was to investigate the relationship between expression of hypoxiainducible faetor-1α (HIF-1α) and uhrastruetural changes in the retina of diabetic rats. Methods 132 SD rats were randomly divided into two groups:control group and diabetic retinopathy (DR) group. Diabetes was induced by intraperitoneal injection of streptozotoein(STZ) in DR group. The techniques of immunohistoehemieal and reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of HIF-1α protein and mRNA in retina respectively at 1,3,6 months after STZ injection ,and retinal ultrastructure was examined by transmission electron microscopy. Results The result of immunohistochemical and RT-PCR revealed little expression of HIF-1α protein and mRNA in retina and the expression level had not significant difference upon time in control group rats. But a considerable increasing expression was seen with the increase diabetes duration in DR group( F = 357. 414 ,P = 0. 001 ). The positive rate of HIF-1α protein was 10. 50% in 1-month diabetic rats and increased to 35.30% in 3-month diabetic rats and to 88.60% in 6-month diabetic rats. No obvious change of retinal ultrastructure was found in the control group. However, swelling of vascular endothelial cell, uneven thickeness or obviously split of basement membrane in retina capillary were found in 3-month and 6-month diabetic rats under the transmission electron microscope. Conclusion Abnormal expression of HIF-1α in retina and retinal uhrastructural changes are closely related to the retinal hypoxia status in diabetic rats,and HIF-1α is involved in the pathogenesis of DR.
