摘 要:目的 研究人类真核翻译延长因子1A2(EEF1A2)及其表达产物eEF1A2蛋白在人胰腺癌细胞株和组织标本中的表达。方法 收集人胰腺导管腺癌病理标本15例,人正常胰腺组织标本8例。运用RT—PCR、Realtime—PCR、Westernblot以及免疫组化等技术检测人胰腺癌细胞株SW1990、BxPC3、PaTu8988和胰腺癌组织标本中EEF1A2的表达水平。结果 人胰腺腺癌细胞株PaTu8988、SW1990和BxPC3中,EEF1A2 mRNA和eEF1A2蛋白表达水平与正常胰腺组织相比.均明显上调。8例癌旁正常胰腺组织标本中仅1例胰腺导管细胞内eEF1A2呈弱阳性表达,而15例胰腺导管腺癌病理标本中.胰腺导管腺癌细胞内eEF1A2均呈中至强阳性表达,两者比较有显著差异(P〈0.05)。结论 EEF1A2在胰腺癌中呈异常高表达。EEF1A2的致癌机制可能与加速蛋白质翻译速度、抑制凋亡以及改变细胞骨架有关。EEF1A2有可能成为胰腺癌一个有效的诊断标志物和基因治疗靶点。Objective Homo sapiens eukaryotic translation elongation factor 1 alpha 2 (EEF1A2) is normally expressed only in the brain, heart and skeletal muscles. However, EEF1A2 was recently shown to be a potential oncogene that is over expressed in ovarian cancer and breast cancer. Pancreatic cancer is the most lethal of major cancers. The molecular mechanisms underlying pancreatic cancer are not well understood. The aim of this study is to elucidate whether EEF1A2 expression might be upregulated in pancreatic cancer. Methods EEF1A2 gene expression in pancreatic cancer cell lines SW1990, BxPC3, and normal pancreas tissues were studied by RT-PCR, Realtime-PCR and Western blots. 15 specimens of pancreatic ductal adenocarcinoma, and 8 specimens of normal pancreas were used to analyze EEF1A2 expression by immunohistochemistry. Results Compared with normal pancreas tissues, EEF1A2 mRNA and eEF1A2 proteins were highly expressed in pancreatic cancer cell lines SW1990 and BxPC3. By immunohistochemistry, although eEF1A2 was barely detectable in the ductal cells of the normal pancreas specimens (1/8), it was moderately to strongly expressed in all of the pancreatic ductal adenocarcinoma specimens (15/15, P 〈 0. 05). Conclusions The above results indicated that EEF1A2 is selectively highly expressed in pancreatic carcinoma. The oncogenicity of EEF1A2 may be related to its role in protein synthesis or to its potential non-canonical functions in cytoskeletal remodeling or apoptosis. Further studies are going to prove whether EEF1A2 is a putative oncogene in pancreas cancer and whether it can be used as a diagnostic marker and therapeutic larget for pancreas tumors.
