摘　　要：目的:观察不同浓度的染料木黄酮和顺铂对耐药卵巢癌细胞系SKOV-3中survivin表达的影响。方法:采用RT-PCR和免疫细胞化学方法,检测不同浓度顺铂和染料木黄酮作用于SKOV-3细胞后survivin mRNA及蛋白表达的变化。结果:与对照组相比,顺铂组survivin mRNA及蛋白表达增强,染料木黄酮组及联合用药组survivin mRNA及蛋白表达减弱(P<0.05)。结论:染料木黄酮能够降低SKOV-3细胞中survivin mRNA及蛋白的表达,并抑制顺铂诱导的survivin过高表达。这一作用可能是其增加SKOV-3细胞对顺铂敏感性的重要机制之一。Objective：To study the effects of genistein and eisplatin at different concentrations on the expression of survivin in drug-resistant human ovarian cancer cell line SKOV-3. Methods： Reversed transeriptase polymerase chain reaction （RT-PCR） and immunoeytoehemieal method were used to detect the mRNA and protein expression of survivin in SKOV-3 cells after treatment with different concentrations of genistein and eisplatin. Results： Compared with the control group,the expression of mRNA and protein of survivin increased in SKOV-3 cells after eisplatin treatment but decreased in cells after genistein or genistein plus cisplatin treatment（P〈0.05）. Conclusion： Genistein can reduce the mRNA and protein expressions of survivin and inhibit the over-expression of survivin induced by cisplatin in SKOV-3 cells. This may be one of the key mechanisms by which genistein sensitizes SKOV-3 cells to cisplatin.
