摘 要:目的:观察db/db自发性糖尿病小鼠颌下腺的病理形态学改变,增殖细胞核抗原(PCNA)在糖尿病小鼠颌下腺中的表达。方法:选取3、4、6、8、10月龄db/db糖尿病小鼠及相应月龄的db/+m正常小鼠颌下腺,应用HE染色及免疫组织化学方法染色后进行图象分析,统计PCNA在颌下腺组织内表达的细胞阳性率。结果:随着糖尿病的发展,颌下腺组织出现腺体萎缩及及颗粒曲管数目减少,实质细胞排列不整齐,呈簇状堆集,结缔组织、纤维及血管增多。PCNA在对照组及糖尿病组颌下腺中均有表达,对照组PCNA细胞阳性率高于糖尿病组。糖尿病组PCNA细胞阳性率随病程延长呈减弱趋势,且减少明显快于对照组。结论:①db/db糖尿病可导致颌下腺组织萎缩及实质细胞形态学改变。②PCNA表达逐渐减弱,较对照组明显降低,说明糖尿病可能导致颌下腺腺体增殖活动的减弱。Objective:To observe the expression of PCNA and morphological changes of submandibular gland of db/db mutant diabetic mice. Methods:Fifty C57BL/Ksj-db/+ m mice were randomly divided into 2 groups, db/db diabetic group and control group. The morphological changes of submandibular gland and the expression of PCNA were observed by using light microscope (LM) and immunohistochemistry. Results:Submandibular gland atrophy and the significant morphological changes in acini and ducts were observed in db/db diabetic mice. PCNA was expressed in submandibular gland in both diabetic mice and control mice. The percentage of PCNA positive cells was significantly decreased in diabetic mice than that in control(P 〈0.01 ), and showed a descend tendency as disease progressed in diabetic group. Conclusion:db/db diabetes mellitus may induce submandibular gland atrophy and inhibit the proliferation of submandibular gland cells.
