盐生杜氏藻碱性蛋白的研究

摘  要:以一种单细胞的盐生杜氏藻为实验材料,利用化学和机械的方法破碎细胞,然后梯度分离细胞核,用盐酸抽提细胞核蛋白质,并与小牛胸腺组蛋白相比较。用SDS-PAGE分析测定所得的碱性蛋白发现盐生杜氏藻核中有三条碱性蛋白带,两条蛋白带与H3和H4相对应,另一条分子量很小。这三条带的表观分于量为14,800,11,700和7.000道尔顿。结果表明盐生杜氏藻的核不具备哺乳动物细胞的五种组蛋白。Cells of Dunaliella salina, a marine alga, were cultured and collected. Cells were lysed with chemical and mechanical methods, and nuclei was prepared. The basic proteins of Dunaliella salina, were obtained with 0.25N HCl extraction of nuclei, and analyzed with SDS-PAGE. Results showed that basic proteins of nuclei of Dunaliella salina, have 2 components corresponding to histones H3, H4 of high mammals, and another band is a protein with small molecule wight. Their apparent molecular weight are 14,800, 11,700, and 7,000dations.
