摘  要:目的:研究神经干细胞(NSC)和脑源性神经营养因子(BDNF)联合治疗对痴呆模型鼠基底前脑p75表达量的影响及其与学习记忆的关系。方法:模拟阿尔茨海默病(AD)大鼠模型,基底前脑注射NSC,侧脑室注射BDNF;Y迷宫测试,免疫组化结合图像分析观察基底前脑p75蛋白表达,半定量RT-PCR法分析基底前脑p75 mRNA表达水平。结果:p75蛋白和p75 mRNA在内侧隔核和斜角带的表达损伤组大鼠较正常组明显下降;移植组较损伤组有明显改善;联合组与正常组无显著性差异。大鼠的学习记忆能力与基底前脑p75 mRNA表达量呈正相关。结论:NSC和BDNF联用较单独使用其一更好地提高p75mRNA表达量和改善痴呆鼠的学习记忆能力。Objective: To study the effects of combined application of the brain derived neurotrophic factor (BDNF) and the neural stem cells (NSCs) on p75 expression in the basal forebrain and the ability of leaning and memory in rats with Alzheimer's disease. Methods: NSCs were transplanted into the transected basal forebrain, and BDNF was injected into the lateral ventricle. The ability of learning and memory was measured by Y-maze test. The level of p75 protein expression in basal forebrain was analyzed by using immunohistochemical method combined with technique of micromeasure and image analysis. RT-PCR method was also used to investigate the expression level of p75 mRNA in the basal forebrain. Results: The expression of p75 protein and p75 mRNA in medial septum (MS) and vertical diagonal branch (VDB) was decreased in injured groups compared with normal control groups; The expression in NSCs groups had more significant improvement than that in injured groups; And no significant difference was found between the combination groups and normal control groups. The ability of learning and memory were positively related to the level of p75 mRNA expression. Conclusion: Combined treatment of NSCs and BDNF is more effective in improving the expression of p75 mRNA and the ability of spatial learning and memory in model rats with Alzheimer's disease than that of the groups treated with BDNF or NSCs alone.
