摘 要:目的研究大鼠实验性脑梗死周边区(A区)星形胶质细胞与突触的关系及其对运动功能恢复的意义。方法经大脑中动脉闭塞制成脑梗死模型后,随机分90只大鼠为抑制、盐水和对照组,以横木行走试验评定运动功能,每组在7、21、42d应用电镜和免疫组织化学方法观察各组大鼠A区星形胶质细胞、突触、胶质纤维酸性蛋白(GFAP)和突触素(SYP)表达值等的变化。结果在3、6周末,抑制组大鼠运动功能、A区突触和星形胶质细胞数目及GFAP和SYP表达值均明显较盐水和对照组差(P<0.01)。结论脑梗死大鼠A区星形胶质细胞可促进突触数目增多、功能增强,这可能是其运动功能恢复的重要机制。Objective To investigate the relation between astrocytes and synapse structure in the region around the infarction area (area A) and the roles of the astrocytes on the recovery of motor skill in the experimental rats with acute cerebral infarction. Methods Respectively,after 7,21.42 d of being made into experimental infarction models by middle cerebral artery occlusion (MACO), 90 rats were divided into 3 group at random: inhibitory, saline and control group. The motor skill of the rats was examined via beam walking test (BWT);and the astrocytes and synapse,the glial fibrillary acidic protein (GFAP), synaptophysin (SYP) and growth associated protein 43(GAP-43) were observed by electron microscope and immunohistochemistry staining respectively. Results After the end of 3 weeks, the recovery of motor skill in the inhibitory was noted less significantly than those in the control or the saline group (P〈0. 01 ). In area A, by electron microscope, the synapse number in the inhibitory increased gradually with the growth of astrocyte, and at 6 weeks their number increased much more significantly than that in the control or the saline group (P〈0. 01 ). By immunohistochemistry staining, comparing the inhibitory with the control, the optical density of GFAP and SYP immunohistochemistry in area A did not show difference significantly at the end of 1 week; Since then, after a gradual increase, the much higher results were got at the sixth weekend (P〈0. 01 ). Conclusions The increase of astrocyte number and vivid is greatly associated with the changes of synapse number and function in area A, which may play an important role on the recovery of motor skill in the experimental rats With acute cerebral infarction.
