乳腺癌病人nm23蛋白的检测与癌转移的关系研究

摘　　要：ｎｍ２３基因是一个与癌转移相关的肿瘤抑制基因，位于１７号染色体着丝点附近，其表达产物为１７ＫＤ的核苷二磷酸激酶。近年来报告，当癌发生转移时，病人的ｎｍ２３蛋白水平下降或检测不出ｎｍ２３蛋白。本文应用斑点ＥＬＩＳＡ法检测了３１例临床病理和Ｘ线均诊断为乳腺癌的病人ｎｍ２３基因的表达水平。检查结果发现，３１例乳腺癌中，２０例阳性，１１例未测到ｎｍ２３基因的表达。经本法检查与临床病理和Ｘ线诊断比较，斑点ＥＬＩＳＡ法检查未发现ｎｍ２３基因表达的１１例中有８例临床病理和Ｘ线诊断证实乳腺癌已转移，两者符合率为７２％。实验结果表明：ｎｍ２３斑点ＥＬＩＳＡ法检查乳腺癌转移，由于方法简便、快速、取材容易，可作为临床判断乳腺癌病人转移的一个生化检测指标。The nm23 gene is a tumor suppressor gene relating with the metastasis of cancer.Near the centrmere of the chromosome 17 with an expressing product about 17 KD named NDP kinase.After having screened the expressing level of the nm23 gene in 31 patients with breast cancer diagnosed by clinical and X ray examination with the method of blot ELISA.We found that expresing product in 20 of them,but no in another 11(however,8 out of them were found to present the metastasis of the breast cancer by clinical and X ray examination).That is to say the concordance is 72%after a careful comparation between these method of blot ELISA is a very simple,quick and the material is also easily accessible to diagnose the transfer of the patients with breast cancer.
