摘 要:目的评价膀胱无细胞基质移植物(BAMG)作为组织工程学细胞载体的可行性。方法10只兔行半膀胱切除术,切取的一半膀胱制备BAMG。另选择20只兔行半膀胱切除术后,用BAMG重建兔膀胱。测定术前和重建术后8周时的膀胱容量、压力和顺应性。HE染色和透射电镜观察BAMG以及重建术后8、16周时的兔膀胱。结果BAMG中未见细胞成分。重建兔膀胱8周时,BAMG中除多层移行上皮外,其余组分还不成熟。16周时,重建膀胱和正常膀胱组织已无法区别。重建膀胱术后8周与术前比较,膀胱的容量、压力和顺应性无显著差异(P> 0.05)。结论BAMG是组织工程学技术中理想的细胞载体。Objective To evaluate the feasibility of bladder acellular matrix graft (BAMG) used as cell carrier for tissue engineering in rabbits. Methods Hemicystectomies were performed in ten rabbits to prepare BAMG. The bladder defects of the other twenty rabbits treated with hemicystectomy were repaired with the BAMG. The rabbits underwent postoperative assessments for bladder functions at week 8, including cystometry, vesical volume, and vesical compliance. The preoperative BAMG and the allografts at week 8 and 16 after surgery were examined by light and electron microscopes. Results No cells could be found in the BAMG. Eight weeks after grafting,all cells in the BAMG were immature except the stratified epithelium.After 16 weeks,it was difficult to distinguish the junction between the host bladder and BAMG by histology. There was no significant difference between urodynamic parameters of the rabbits before surgery and at week 8 after surgery. Conclusion BAMG is the ideal cell carrier for tissue engineering.
