摘 要:目的探讨雌激素受体在骨质疏松中的作用及雷洛昔芬对其调整作用。方法选择3月龄Sprague-Daw- ley雌性大鼠48只,随机分为假手术组、去卵巢组、雌激素组及雷洛昔芬组,每组12只,给药3个月后处死大鼠。分离获取股骨,应用RT-PCR技术检测骨细胞中ER mRNA表达水平。结果雷洛昔芬使去卵巢大鼠骨密度增加,提高ERαmRNA,对ERβmRNA的表达无明显的影响作用。结论雷洛昔芬可能通过对ER基因表达的影响而起到治疗骨质疏松的作用。Objective To explore the effect of estrogen receptor in osteoporosis and the effect of raloxifene on estrogen receptor expression of bone in oophrectomy rots.Methods A total of 48 female Sprague-Dawley rats were divided into 4 groups:the sham-operated group(12 rats) ,the ovariectomized group(12 rats) ,the estrogen group(12 rots) and the raloxifene group(12 rats).After 3 months treatment, all rats were killed,the uterus was got for histomorphological analysis and the bone were excised and examined using RT-PCR. Results The bone desity increased after raloxifene treatment, the expression of ERα mRNA elevated, ERβ mRNA showed no significant increase. Conclusion Raloxifene may play an important role in bone metabolism by affecting the expression of estrogen receptor and has no proliferation effect on endometrium.
