摘 要:目的观察两种ω-3多不饱和脂肪酸(ω-3polyunsaturated fatty acid,ω-3PUFA)二十碳五烯酸(eicosapentae-noic acid,EPA)和二十二碳六烯酸(docosahexaenoic acid,DHA)对高转移性人前列腺癌细胞PC-3体外黏附和侵袭能力的影响,并探讨其相关分子机制。方法MTT法观察EPA和DHA对PC-3细胞增殖的影响;以体外黏附和侵袭实验观察EPA和DHA对肿瘤细胞黏附和侵袭能力的影响;RT-PCR法观察EPA和DHA对与细胞骨架相关的RhoA、Rac1、Rac2和Cdc42基因mRNA表达的影响。结果EPA及DHA均能抑制PC-3细胞的增殖,并呈剂量效应关系;PC-3细胞以60μmol/L EPA和DHA分别处理后,体外黏附和侵袭能力均降低,在matrigel基质胶上黏附30、60、90min和120min的黏附率均低于对照组(P<0.05,P<0.01),侵袭18h后的抑制率分别为(35.65±7.76)%和(41.32±5.86)%(P<0.01),RhoA、Rac2和Cdc42基因mRNA表达均不同程度下降。结论ω-3PUFA可能通过下调Rho GTP酶mRNA的表达,抑制PC-3细胞体外黏附和侵袭能力。Objective To investigate the effects of 2 kinds of ω-3 polyunsaturated fatty acid, eicosapentaenoic acid (EPA) and docosahexaenoic acid ( DHA), on the adhesion and invasion of PC-3, a high metastatic human prostate tumor cell line, and explore the possible mechanism. Methods MTF assay was used to determine the effects of EPA or DHA on the proliferation of PC-3 cells. Adhesion assay and invasion assay were used to observe the anti-adhesion and anti-invasion effects of EPA or DHA on PC-3 cells, respectively. RT-PCR analysis was conducted to detect the expression of RhoA, Rac1, Rac2 and Cdc42 mRNAs. Results Both EPA and DHA inhibited the growth of PC-3 cells in a dose-dependent manner. After treated with EPA or DHA of 60 μmol/L, the adhesion and invasion abilities of the cells were decreased. The adhesion rate were lesser than that of the control when adhered for 30, 60, 90 and 120 min on matrigel, and the invasion inhibitory rate were (35.65 ±7.76)% in EPA treated group and (41.32 ± 5.86)% in DHA treated group (P 〈0. 01 ). The expressions of RhoA, Rac2 and Cdc42 in PC-3 cells were all decreased in compare with those of the control. Conclusion These results suggest that ω-3 PUFA can inhibit the adhesion and invasion of PC-3 ceils by down-regulating the expression of the Rho GTPase.
