摘  要:背景:炎症性肠病(IBD)的发病与一系列免疫异常有关。免疫反应受特定的免疫调节细胞调控,包括CD4+CD25+T细胞,而CD4+CD25+T细胞的发育和功能可能受FOXP3基因调控。目的:研究FOXP3的表达特点以及IBD时FOXP3表达的改变。方法:以免疫磁珠分离CD4+、CD4+CD25+和CD4+CD25-T细胞。以逆转录聚合酶链反应(RT-PCR)和蛋白质印迹法检测IBD患者和非IBD对照外周血单个核细胞(PBMC)、肠固有层单个核细胞(LPMC)和肠黏膜组织中FOXP3的表达。结果:CD4+CD25+T细胞中FOXP3mRNA的表达明显强于CD4+T细胞和未分离的PBMC或LPMC。T细胞受体(TCR)激活后,或经白细胞介素(IL)-2、IL-15作用后,PBMC或LPMCFOXP3mRNA或蛋白表达明显增强。12例IBD中9例(75.0%)肠黏膜LPMC中检测到FOXP3mRNA表达,高于对照肠黏膜的47.1%(8/17,P>0.05)。未受刺激的IBD和对照肠黏膜组织中未检测到FOXP3mRNA表达。结论:人类FOXP3的表达受T细胞活化影响。IBD患者肠黏膜LPMC中FOXP3mRNA的表达率有增高的趋势,但与对照肠黏膜相比无显著差异。Background: The pathogenesis of inflammatory bowel disease (IBD) is associated with a variety of immunologic abnormalities. The immune response is modulated by distinct types of immunoregnlatory cells, including CD4^+CD25^+ Tcells whose development and function may be controlled by the FOXP3 gene. Aims: To investigate the characteristics of FOXP3 expression and possible abnormalities of FOXP3 expression in IBD. Methods: CD4^+, CD4^+CD25^+ and CD4^+CD25^- T-cells were isolated by immunomagnetic beads. The expression of FOXP3 in peripheral blood mononuclear cells (PBMC), intestinal lamina propria mononuclear cells (LPMC) and intestinal mucosa of IBD patients and non-IBD controls was assessed by reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot. Results: FOXP3 mRNA expression was much stronger in CD4^+CD25^+ T-cells than that in CD4^+ T-cells and unseparated PBMC and LPMC. Its expression was clearly upregulated in both PBMC and LPMC by the engagement of CD3 receptors, or stimulation with interleukin (IL)-2 and IL-15. FOXP3 mRNA expression was detected in 9 of 12 IBD and in 8 of 17 control LPMC (75.0% vs. 47.1%, P〉0.05). No FOXP3 mRNA expression was detected in unstimulated IBD or control mucosa. Conclusions: FOXP3 expression in human cells is affected by T-cells activation. LPMC of IBD tends to express increased FOXP3 mRNA, but there is no significant difference between IBD patients and the controls.
