β-防御素-2在小鼠噁唑酮结肠炎黏膜中的表达及其意义

摘　　要：背景:炎症性肠病(IBD)的病因和发病机制尚不完全清楚。最近研究发现其可能与天然免疫效应分子防御素异常有关。目的:探讨结肠炎黏膜中β-防御素-2的表达与促炎细胞因子肿瘤坏死因子(TNF)-α和白细胞介素(IL)-1β表达的关系。方法:建立BALB/c小鼠噁唑酮结肠炎模型,并将其分为正常对照组和噁唑酮结肠炎组,行大体和组织学评分,免疫组化、实时荧光定量逆转录聚合酶链反应(RT-PCR)检测结肠黏膜中β-防御素-2和促炎细胞因子TNF-α、IL-1β蛋白及其mRNA的表达。结果:噁唑酮结肠炎组结肠黏膜可见明显的肉眼和组织学炎症,β-防御素-2的表达显著高于正常对照组,与结肠炎症程度呈正相关。β-防御素-2蛋白及其mRNA表达与TNF-α和IL-1β蛋白及其mRNA的表达呈正相关。结论:β-防御素-2的表达与促炎细胞因子表达密切相关,两者相加可能是结肠炎症反应放大、持续迁延的原因之一。Background： The etiology and pathogenesis of inflammatory bowel disease （IBD） are still not well-known. Recently, it has been speculated that IBD originates from an alteration in mucosal defensins. Aims： To explore the correlation between expression of β-defensin-2 and pro-inflammatory cytokine, such as tumor necrosis factor （TNF）-α and interleukin （IL）-1β in colitis mucosa. Methods： Colitis induced by oxazolone was constructed in BALB/c mice, which were divided into two groups： control group and oxazolone colitis group. The macroscopic and histological score of colon were evaluated. Expression of β-defensin-2, TNF-α and IL-1β and their mRNA in colonic mucosa were detected by immunohistochemistry and real-time fluorescent quantitative reverse transcriptase polymerase chain reaction （RT-PCR）. Results： Oxazolone colitis was constructed successfully with marked inflammation in colonic mucosa examined macroscopically and histologically. Expression of β-defensin-2 in colonic mucosa was significantly higher than that in control group, moreover, expression of β-defensin-2 and its mRNA are positively correlated with expressions of TNF-α and IL-1β, and their mRNA in colonic mucosa of oxazolone colitis group. Conclusions： Expressions of β-defensin-2, TNF-α and IL-1β are closely correlated, which may play an important role in magnifying and sustaining inflammation of the colon in IBD.
