摘  要:通过重组幼仓鼠肾细胞(BHK细胞)在不同温度下的批培养和脉冲培养,研究了温度对重组BHK细胞生长、代谢以及血管性血友病因子(vWF)蛋白表达的影响。相对于37℃,升高和降低温度的培养都降低了细胞的生长速率和密度,降温的作用更明显,主要表现为延长了细胞生长的迟滞期。在不同温度的脉冲培养中,细胞的比生长速率变化不大,表明进入对数生长期后,细胞生长对一定范围的温度变化不敏感。培养温度对细胞群体中处于S期的细胞比例影响不大,但在33℃和39℃下培养,细胞群体中处于G0/G1期的细胞比例有明显升高,处于G2/M期的细胞比例下降。当培养温度降低到33℃时,细胞群体中发生凋亡的细胞数随着温度升高而增加。温度的降低能显著提高单个细胞的vWF蛋白表达能力,在33℃下培养,vWF平均比生产速率提高了45%。Effects of temperature on the growth, metabolism and vWF expression of recombinant baby hamster kidney (rBHK) cells were investigated in batch and pulse cultures. It was demonstrated that both higher and lower temperatures could retard rBHK cell growth compared with usual 37 ℃, more severe under lower temperature. The growth retardation by temperature change is attributed mainly to the prolonging of lag phase, and the specific growth rate of rBHK cells was almost independent on the variation of temperature in exponential phase. Temperature change had little effect on the cell percentage in S phase of cell cycle, but the cell percentage increased in G0/G1 phase and decreased in G2/M phase obviously. Moreover, the apoptotic cell number of cell population increased with the rising of temperature. The lowering of temperature could remarkably increase vWF expression per cell, the specific production rate of vWF increased by 45 % at 33 ℃ compared with 37 ℃.
