摘 要:目的以活体检测探讨帕金森病(PD)猴模型神经递质含量的改变。方法应用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)制备偏侧PD猴模型,采用颅内微透析技术结合高效液相色谱-电化学方法(HPLC-EC)活体检测双侧尾核区多巴胺(DA)及其代谢产物的含量。结果MPTP注射侧与注射对侧相比,尾核区DA、3,4-二羟基苯乙酸(DOPAC)和高香草酸(HVA)的含量分别减少70%、34%和38%。结论颅内微透析技术是活体检测脑内神经递质含量的最有效方法。Objective To measure the extracelluar transmitters in monkey model of Parkinsonism in vivo.Methods Unilateral intracarotid infusion of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) produces a useful monkey model of hemiparkinsonism. Brain microdialysis,combined with high performance liquid chromatography and electrochemcal detection (HPLC-ECD), was employed to measure the levels of dopamine(DA)and its metabolites in the bilateral caudate nuclei of hemiparkinsonian monkey. Results The levels of DA,DOPAC and HVA in MPTP-treated side were decreased by 70%, 34% and 38% respectively,as compared with those in the untreated side. Conclusions The result suggests that brain microdialysis is a valuable tool for measuring in vivo neurochernical changes,and it helps to comprehend the relationship between symptoms of Parkinsonism and changes of the extracelluar neurotransmitters.
