摘  要:目的:探讨放疗对舌鳞癌Tca 8113细胞和耐药的Tca 8113/CBDEA细胞耐药性的影响。方法:应用免疫组化SP法检测放疗对P-糖蛋白(P-glycoprotein,P-gp)、多药耐药相关蛋白1(multidrug resistance associate pro-tein 1,MRP1)和谷胱甘肽硫-转移酶π(glutathione S-tranferaseπ,GST-π)表达的影响和检测放疗对细胞内阿霉素浓度的影响。结果:Tca 8113/CBDEA细胞在放疗后4 h P-gp,MRP1,GST-π耐药蛋白表达无明显上升,24 h耐药蛋白的表达明显升高(P<0.01),Tca 8113细胞的耐药蛋白表达在4 h和24 h时均有明显上升(P<0.01)。放疗以后肿瘤细胞内阿霉素(ADM)浓度有明显下降。结论:放疗可引起舌鳞癌细胞的耐药。提醒我们对舌癌患者进行放疗和化疗的方案设计时应考虑此点。Objective: To study effect of radiotherapy on expressions of multidrug resistance(MDR) factors and intracellular ADM concentration in tongue squamous cell carcinoma cell line Tca 8113 and its MDR cell line Tca8113/CBDEA. Methers: Cell lines were treated with 2Gy of γ- radiation for 4hs and 24hs. Immunohistochemistry was used to detect expressions of MDR relative proteins ( P - gp, MRP1, GST - π) and HTS 7000 Plus Bio Assay Reader was used to measure intracellular ADM concentration. Results: After radiotherapy, there was no significant increase of MDR relative proteins in Tca 8113/CBDEA cell line 4h. But after 24h, it significantly increased (P 〈0.01 ). Expressions of MDR relative proteins in Tca 8113 significantly increased at 4h and 24h postradiotherapy (P 〈0.01 ). Intracellular drug concentration decreased significantly at 4h and 24h postradiotherapy in the cell lines (P 〈0.01 ). Conclusion: MDR of tongue squamous cell carcinoma cell line can be induced by radiotherapy. The results might explain the poor efficacy of chemotherapy following radiotherapy and contribute to consideration of new treatment strategies in the management of tongue squamous cell carcinoma.
